RolesofGABABreceptorsinthespinaldorsalhornofratswithdiabeticneuropathicpainABSTRACTObjective:Aswekonw,theneuropathiesareoneofthemostcommonlong-timecomplicationsofdiabetes,andthemorbilityincreseseveryyear.About16%and26%ofdiabeticpatientssufferchronicpain.Thismaybereferredtoasdiabeticneuropathicpainorpainfuldiabeticneuropathy.Themechanismsofdiabeticneuropathicpainareverycomplexandthemanagementisstillquitedifficult.Studiesonthetransmitterphmaraeologyhaveindicatedthatthereexistedchangesofneurotransmittersandreceptorsinspinalcordinducedbyneuropathicpain.ChangesofGABABreceptorscouldaffectthefunctionofGABAergicsystemsothattoaffectthetransmissionandmodulationofnociceptivemessages.ThepersentstudyistoexplorechangesofGABABreceptorsinspinalcorddorsalhornofratswithdiabeticneuropathicpaininducedbySTZ.Characteristicbehavioralchangeswereobservedtoaffecthyperalgesiaofrats,andwealsohadintrathecalinjectedbaclofentoobservethechangeof50%pawwithdrawthreshold(PWT)ofratswithdiabeticneuropathicpain.Methodsofimmunohistochemistry,RT-PCRandwesternblottingwereappliedtoexaminetheexpressionofGABABreceptorsinspinalcorddorsalhorn.StudiesaboutthechangesofGABABreceptorsinspinalcorddorsalhorncouldhelpfindingoutmorespecifictargetsoftreatmentfordiabeticneuropathicpain.Methods:100maleSprague-Dawleyratswererandomlyselectedforthisexperiment,weighing130-150g.Bodyweights,fastingbloodglucoseleveland50%pawwithdrawthresholdofalltheratsweremeasuredbeforeSTZinjection.90ratswereinducedwithasingleintraperitonealinjectionofstreptozocin(50mg/kg)asdiabetes.AtoneweekafterSTZinjection,74ratswhosefastingbloodglucoselevelincreasedupto16.7mmol/Lwereconsidedasdiabeticrats,andtheratiois82%.AttwoweekafterSTZinjection,responsestothemechanicalstimulusofdiabeticratsweremeasuredwithvonFreyfilament,andwhose50%pawwithdrawthreshold(PWT)weredownto4gwereconsidedasdiabeticneuropathicpainmodels.Another10ratswerewereintraperitonealinjectedofsalineascontrols.30diabeticratswererandomlyselectedforbehavioarlexperiment.23didnotappearnerveinju
