ABSTRACTOwingtobiologicalaeratedfilter(BAF)reactorhadbetternitrogen removal efficiency,this paper studied on separation, identification andapplicationofammonifyingbacteriafromBAFreactor.Firstly,9 ammonifyingbacteriumwereinitiallyscreenedfromthebiofilmoftheBAF reactor.ThenweelectedthestrainT03andT05whichcouldeffectively decomposeorganicnitrogenfromthe9strainsforfurtherstudiesonthe ammoniationcharacteristics.What’smore,throughmorphologicobservation andphysiological-biochemistrycharacteristicanalysis,strainsT03,T05were preliminarilyidentifiedasAlcaligenesandMicrococcusrespectivelywhich referedto“IdentificationManualofCommonBacterialSystems”.Inaddition, theresearchofT03,T05forammoniationcharacteristicsindicatedthatthe strains were aerobic, the best nitrogen source was protein, the suitabletemperaturewas30℃,thesuitablepHvaluewas7.0andthebestinoculumquantitywas5%.Intheseoperatingparameters,organicnitrogeninthe culturecouldbeeffectivelydecomposedbythestrainsandorganicnitrogen decompositionrate wasupto86.91%and69.98%respectively.Whenmicroorganismswereindifferentnutritionalconditionsand environmentalconditions, themyceliumgrowthspeedwas different.The fermentationcompositionandfermentationconditionsofthestrainsT03and T05wouldbeoptimizedbydesigningorthogonaltests.TheresultsshowedthatstrainT03fermentationconcentrationwas10.37g/Lfor24h,at30℃,in7.0pHvalueand5%inoculums.Andingredientofthemediumincluded3.0g glucose,5.0gPeptone,3.0gbeefextract,0.3gK2HPO4,0.25gNaCland0.01g FeSO4 in 1000mL distilled water. And that strain T05 fermentationconcentration was 8.47 g/L for 30h, at 35℃, in 7.0 pH value and 5%inoculums.Andtheingredientofthemediumincluded3.0gglucose,5g Peptone,3.0gbeefextract,0.5gK2HPO4,0.75gNaCl,and0.01gFeSO4 in1000mLdistilledwater.Polymerasechainreaction(PCR)productswereobtainedbythePCR sequencingandmatchingwithMarker.Theelectrophoresiswasasingleclear beltandwasgoodofspecificitywithoutapparentnon-specificamplification products.Thesizeof16SrDNAfragmentswas1426bpbyse
