AbstractObjectiveTo study the therapeutic effect of Aurora B kinase inhibitors、VX-680 and the cisplatin on the nude mice transplants of hepatic carcinoma, and observe the effect to the nude mice transplants of hepatic carcinoma and the chemo-sensitivity.MethodsRandomly divide 24 male nude mice injected with the human hepatoma cell line HepG-2 into 4 groups: combination group, VX-680 group, cisplatin group and control group. Since the 7th day after experimental tumor inoculation, Group VX-680, with the dose of 12.5 mg/kg, intraperitoneal injection for two times one day, 14 times totally; cisplatin, two times every week,40mg/kg, intraperitoneal injection,one week in a row, the 7th and the 14th day each time. The control group was injected with normal saline, 40mg/kg and the duration of a week, then measure the line of apsides of the tumor growth in nude mice and measure the gross tumor volume, in the next day. One week after drug withdrawal, completely remove the tumor mass from the dead mice, study into the impact of tumor and the growth of hepatocellular carcinoma in nude mice, by texting Aurora in combination group, VX-680, cisplatin group and control group, and Bcl-2 B, P53 with Western blot and immunohistochemical method .ResultsVX-680 and cisplatin both were able to make the tumor volume reduce significantly, significant reduction in the combination group (P 0.05). Detected by Western blot and immunohistochemical method, in the combination group Aurora B kinase expression decreased, and in the combination group, Aurora B kinase inhibitors expression decreased, P53 expression increased, Bcl-2 expression decresed more significantly(P0.05) .Conclusion1.Aurora B kinase inhibitors Vx-680 can significantly inhibit the growth of the murine tumor cells of HEPG-2 bearing cancer. 2.When the Aurora B kinase inhibitors VX-680 is combined with the conventional chemotherapy drugs DDP, the combination can in crease the chemo-seasitivity of the hepatoma carcinoma significantly
