Experimental study of the effect by Buyanghuanwu decoction on the optic nerve injury model of rats Abstract Objective: The observation of the influence on the following things by Buyanghuanwu soap is firstly accomplished in this thesis. The first one is the shape of the retina, which is neurologically injured, and the second one is the number of the retinal ganglion cells. The last thing is expression of retinal growth associated protion 43. Based on the result,, the performance and the theory of the Buyanghuanwu soap used for treating optic nerve is discussed. Method: 1. Select 40 female clean-level grown-up SD rats and divide them into four group, including a normal group which includes 5 rats, a fake-surgery group which has 5 rats, a reference group which owns 15 rats and a treatment group in which there are 15 rats. The retinal nerves of the rats in the fake-surgery group are only exposed, not clamped. As for the reference and treatment group, the model of the animal with calmed retinal nerve is built. Then the groups are divided into 3 subgroups, 5-day, 14-day and 28-day subgroups based on the observation time after modeling. There are 5 rats in each of them. 2. On the day after modeling, the rats in the treatment group are fed with Buyanghuanwu soap. Simultaneously, the rats in the reference and fake-surgery groups are fed with equal volume saline. The rats in the normal group are dealt with as usual. 3. Fetch the materials on the day 5, 14, 28 days later than the modeling day. Then the shapes of the retina with injured retinal nerve of the rats are observed under the HE staining. After that, the number of the retinal ganglion cells is investigated by using pathology image analyzer, and the expression of GAP-43 in the retinal ganglion cells is detected by using immumofluorescence method.
