Cy3 is an orange-red fluorescent dye that is particularly good for protein labeling, producing highly fluorescent protein conjugates. The dye is a superior alternative to the commonly used carboxytetramethylrhodamine (TAMRA) because it does not form aggregates and is very water-soluble. CY3 NHS ester is readily coupled to an amine group using the following recommended procedure.
Typical Procedure for Labeling 1mg of IgG proteina
Prepare 1mL of 1 M sodium bicarbonate (NaHCO3) solution by dissolving 84 mg of sodium bicarbonate in 1 mL deionized H2O. The resulting bicarbonate solution has a pH of ~8.3.
If the proteinb is in a solid format, dissolve 1 mg of the protein in 0.5 mL 0.1M NaHCO3 buffer that is made from the 1 M NaHCO3 solution in step 1) by diluting 10 times with deionized H2O.
If the protein is already in a solution and is greater than 2mg/mL, dilute the protein solution with PBS or 0.1 M sodium bicarbonate buffer to 2mg/mL. To 0.5 mL of 2mg/mL protein solution, add 50 ?L of the 1 M bicarbonate solution prepared in step 1.
Prepare a stock solution of the dye by dissolving the dye in anhydrous DMF at 1mg/100?L concentration. The solution should be stable for a few days if kept at 4 (C. Regular DMF may be used for dissolving the dye, but the solution should be used within hours due to potential hydrolysis of the dye.
