人参总皂苷对PC12细胞突起生长及细胞中GAP-43表达的影响 李晗宇1,2 王顺和 1,2 郑娟1,2 姜蓉2 （1.重庆医科大学病理学教研室；2.重庆医科大学干细胞与组织工程研究室;重庆400016） 【摘要】 目的 研究人参总皂苷(Total saponin of Panax ginseng ,TSPG)对PC12细胞突起生长以及神经细胞突起可塑性信号分子GAP-43表达的影响。方法：以0、10、20、40、80mg/L浓度的人参总皂苷体外培养PC12细胞，应用倒置相差显微镜观察细胞形态学改变，透射电镜观察突触连接，显微镜测微尺测量突起长度，免疫组织化学技术、western blot检测生长相关蛋白-43（GAP-43）的表达。结果： 10、20、40、80mg/L浓度的TSPG组GAP-43阳性细胞数明显高于对照组，且以40mg/L组最为明显（P＜O.O5）。与对照组比较，TSPG组分化的细胞其突起和细胞直径明显增长和增大，以40mg/L组最为明显（P＜O.O5）。Western blot检测结果显示，与对照组相比，10、20、40、80mg/L浓度TSPG组细胞GAP-43蛋白水平明显增加，以40mg/L组最为明显（P＜O.O5）。TSPG能促进PC12细胞形成突触连接。结论：TSPG能诱导PC12细胞分化、突起延伸，而且还能促进GAP-43蛋白的表达。 【关键词】 人参总皂苷；PC12细胞；突起生长；GAP-43 【中图分类号】R282.71 Q28 Effect of Total saponin of Panax ginseng(TSPG) on neurite-outgrowth and expression of Growth associated protein 43（GAP-43）on PC12 cells LiHanyu1,2, WangShunhe1,2, ZhengJuan1,2, JiangRong2 (1.Department of pathology;2.Stell cell and organization project laboratory, Chongqing Medical University,Chongqing,400016,China) Abstract: Objectiv: To investigate the effect of Total saponin of Panax ginseng(TSPG) on neurite-outgrowth and expression of GAP-43 on PC12 cells. Methods: Cultured PC12 cells with 0、10、20、40、80mg/L TSPG in vitro, respectively, the morphological changes of PC12 cells were observed by inverted microscope,the synaptic linkage were observed by transmitting electronic microscope, the expression of GAP-43 was detected by immunohistochemical and western blot technique. Results: The number of GAP-43 positive cells was significantly increased in10、20、40、80mg/L TSPG groups compared with contral group and the most significant concentration was 40mg/L(P＜0.05).The length of neurite and max diameter of PC12 cells in TSPG were obviously longer and bigger than those in contral group and the most significant concentration was also 40mg/L(P＜0.05).
