238 眼科 2002 年第 11 卷第 4 期 　Ophthalmol CHN ,2002 ,Vol 11 ,No. 4 视网膜定向铺片方法 马 　科 ,徐 　亮 (北京市眼科研究所 ,北京 100005) 摘 　要 :目的:介绍一种大鼠视网膜定向铺片方法。方法 :对不固定、固定 1～12 小时及固定 24 小时以上 SD 大鼠眼球采 用碳素标记视网膜定向铺片 ,剪成上、下、鼻、颞四个区域。结果 :不固定组视网膜容易撕裂 ,固定24 小时以上组视网膜周边上 卷 ,固定 1～12 小时组视网膜分离容易 ,铺片后视网膜平整并很好辨别各方向。结论 :该方法简单实用 ,能很好地确定铺片后 视网膜的方向 ,对于视网膜节细胞的定量分析等研究有重要意义。 　　关键词 :视网膜 ,铺片 ;动物 ,实验 　　中图分类号 :R774 　　文献标识码 :A 　　文章编号 (2002) Method for direction f inding flattening of rat retina on the slide/ MA Ke …∥Ophthalmol CHN. 2002 ,11 (4) . 238 ～239 (Beijin g Institute of Ophthalmology ,Beijin g 　100005) Abstract :Objective : To introduce a method of direction finding flattening of rat retina on the slide. Method : Eyes of 12 rats ,after labeled with ink on the upper conjunctiva ,were devided into 3 groups ,and the enucleated eyeballs in group Ⅰwere not fixed with any fixation fluid ,while in group Ⅱ,the eyeballs were fixed in 4 % polymer formalin 0. 1M PBS 1～12 hrs. ,and in group Ⅲ,the fixation time was over 24 hrs. According to the conjunctival ink mark ,the rat retinae were cut with scissors into upper ,lower ,nasal and tempo ral pieces ,and flattened on the slide. Results :Of 8 eyes in group Ⅰ,6 eyes were torn easily ,while those fixed for over 24 hrs. ,the edges of retinae rolled up. The retinae fixed for 1～12 hrs ,separated from choroid easily ,and after flattened on the slide ,they were flat and direction can be identified. Conclusion : This method is simple and useful ,by which the direction of rat retina can be identified ,and can be used in quantitative analysis of retina ganglion cells. 　　Key words :retina ,flattening ;animals ,laboratory 　　视网膜平铺片用于多种研究之中 ,包括视网膜 组置于 4 %多聚甲醛 01M PBS 固定 24 小时以上。 逆行标记节细胞定量研究 ,单个节细胞的定位注射 , 在碳素标记相应部位的后
