化疗前后肿瘤组织与PBMC MDR1表达的相关性研究 桂琳玲 金先庆 罗庆 赵珍珍 赵利华 郭振华 杨坤 （重庆医科大学附属儿童医院普外科，重庆400014） 摘要：目的 化疗前后肿瘤组织与外周血单个核细胞MDR1表达的相关性研究。方法 建立小鼠肿瘤耐药模型，MTT法检测化疗前后肿瘤对多种化疗药物耐药指数。半定量PCR法检测化疗前后肿瘤组织和外周血MDR1a mRNA表达量。柔红霉素排出试验证实p-gp外排功能。结果 化疗后肿瘤对ACT、ADM、VCR、HCPT、5-Fu、DAM、VP16的耐药指数分别为29.3、26.4、10.9 、7.0、5.8、3.0、2.8。肿瘤组织和外周血MDR1a mRNA 表达随着化疗时间的延长表达增加，并存在一定相关性（r为0.903）。化疗后肿瘤组织和外周血对柔红霉素的外排能力明显高于未化疗组(p0.05)。结论 肿瘤组织和外周血单个核细胞MDR1表达有一定相关性，检测外周血MDR1表达作为评价临床化疗药物对肿瘤是否耐药的指标具有一定可行性。 关键词：MDR；肿瘤；单个核细胞；半定量PCR 中图分类号：R33 ，R73 文献标志码：A The correlation research of tumorous tissue and PBMC expressive MDR1 before and after chemotherapy GUILin-ling JINXian-qing LUOQing ZHAOZhen-zhen ZHAOLi-hua GUOZhen-hua YANGKun （Department of Surgery ,Chongqing Medical University,Chongqing400014,China） Abstract：Objective The correlation research of tumorous tissue and peripheral blood mononuclear cell(PBMC) expressive MDR1 before and after chemotherapy. Methods Establish the tumor drug resistance model of mouses. MTT detects drug resistance index numbers of the tumor after chemotherapy. Semiquantitative PCR detect MDR1a mRNA of tumor and PBMC before and after chemotherapy.DMA expulsion test checks the expulsive function of p-gp. Results Drug resistance indexs are 29.3、26.4、10.9 、7.0、5.8、3.0、2.8 respective to ACT、ADM、VCR、HCPT、5-Fu、DAM、VP16 after chemotherapy .The tumor and PBMC,MDR1a mRNA expression increase after chemotherapy and have surely correlation (coefficient correlation is 0.903) and p-gp excretive ability is higher than no chemotherapy group. Conclusion The expression of MDR1a mRNA of tumor and PBMC have correlation.The expression MDR1 can become a index as evaluation clinical chemotherapy drug whether drug resistance to tumor.
