Experimental Study on Changes in Tumor Ang-2, VEGF Expressions after Gamma Knife Treatment on C6 Glioma-Bearing Rats Abstract Objective: By building C6 glioma-bearing rats’ after gamma knife treatment model, respectively detect Ang-2, VEGF genes in tumor tissues as well as differences in protein expression levels between treatment group and control group, explore the action mechanism of gamma knife on vascular damage effect in glioma radiotherapeutic action. Methods: (1) cultivate C6 glioma cells through in vitro culture and passage, gather cells in logarithmic phase and prepare cell suspension, use micro injector to slowly inject 5 μL cell suspension into rats’ intra craniums, then seal them with bone wax, feed regularly, 2 weeks later, conduct MRI test, if tumor is found, the model building is successful. Randomly divide the rats into treatment group and control group, conduct gamma knife treatment to the treatment group. (2) 7 days after gamma knife treatment, sacrifice the rats, detect Ang-2, VEGF genes and protein expression levels in tumor tissue using immunohistochemical method, RT-PCR, western-blotting method respectively, and conduct statistical analysis. Results: (1) The results showed that the method of regular feeding after intracranial injection of high concentration C6 glioma cell suspension can make the success rate of tumor-bearing rat modeling over 95%. (2) Drawing materials 7 days after radiotherapy, the immunohistochemical results showed that the treatment group’s positive rate of tumor tissue Ang-2 was 32.83%, which was significantly decreased compared to 70.01% of the control group, the difference was statistically significant (P 0.01); treatment group’s positive rat
