· 1430· 史文荣，等 Canstatin基因导入抑制裸鼠肝癌移植瘤的生长及血管生成 Canstatin基因导入抑制裸鼠肝癌移植瘤的生长及血管生成史文荣，王雪雯，宋海林，段晶晶 Canstatingeneimpoflinhibitsgrowthandangiogenesisoftransplanted livercancerin mice ShiWenrong，WangXuewen，SongHailin，DuanJingjing DepartmentofPathophysiology，MedicalSchoolofShiheziUniversity，XinfiangShihezi832000，China．【Abstract】 Objective：ToinvestigatetheeffectofCanstatingenetherapyongrowthandangiogenesisoftransplan— tedHepG2livercancerinnudemice．M ethods：TransplantedHepG2livercancermousemodelwasestablished，using Ad——Canstatin／GFPgenetherapy．TheexpressionofCanstatinproteinintransfectedtumorsweredetectedbyWestern blotting．TherapeuticeffectofAd—Canstatin／GFPontransplantedHepG2livercancerwasobserved．Thedamageof kidneyandliverwereobservedbyHEdyeing，andthenumberofWBC inbloodwascounted．Microvasculardensity andvascularmimicryformationoftransplantedtumorsindifferenttherapygroupswasobservedbyimmunohistochem— istry．Results：Intotumorratewas100％，andAd—Canstatingroupofnudemouseappetitewasgreaterthanthetwo controlrgoup (P0．05)．Drinkingwaterandexcretionwaslessthanthetwocontrolgroup (P0．05)．Weight growthcurvedisplayedthatAd—Canstatingroupwasincreased，huttwocontrolgroupwasnotincreaseordecrease． ThetumorvolumeofAd—Canstatingrouprgowthmoreslowlythantwocontrolgroup，andthedifferenceintreatment ofthefirst10days(P0．05)．ThetumorweightofAd—Canstatingrouplessthantwocontrolgroup(P0．05)， andinhibitingtumorrate：37．50％．InAd—Canstatingroupliverkidneyhematopoieticsystem werenouma1．Thenum— betofMVD，VM inAd—Canstatingroupwerelessthantwocontrolgroup(P0．05)．Conclusion：Adenovimsme— diatedCanstatingenecan inhibittumorangiogenesis，thusinhibitingtransplantedHepG2livercancertumorgrowth no ， sideeffectsintheexperiment．
