 黄芩素对人皮肤鳞状细胞癌细胞系 SCL-1 增殖抑制及诱导凋亡作用的研究 # 5 10 15 20 25 30 35 40  卢军，刘梅，何春涤** （中国医科大学附属第一医院皮肤科 辽宁 沈阳 110001） 摘要：目的：本实验应用黄芩素处理人皮肤鳞状细胞癌 SCL-1 细胞，观察黄芩素对人皮肤 鳞癌细胞生长的作用。方法：先对 SCL-1 细胞和 HaCaT 细胞进行培养，用四甲基偶氮唑蓝 （MTT）法检测细胞增殖抑制率，在倒置显微镜下观察 SCL-1 细胞形态，流式细胞仪测定 细胞早期凋亡，并用丫啶橙荧光染色观察细胞凋亡后的形态学变化。结果：不同时间组两两 比较，黄芩素处理后 48h 与 24h，12h 的组间的差异有统计学意义，用 50μ g/ml 黄芩素溶液 作用 SCL-1 细胞 24h 后，SCL-1 细胞出现了凋亡现象。结论：黄芩素对 SCL-1 细胞的增殖 有抑制作用，随着作用时间的延长抑制效果更明显，并可以诱导 SCL-1 细胞发生凋亡。黄 芩素对于 HaCaT 细胞的生长抑制作用不明显；全反式维甲酸也可以抑制 SCL-1 细胞的增殖； 一定浓度的黄芩素与全反式维甲酸联合作用对于 SCL-1 细胞的抑制增殖作用有协同效应。 关键词：SCL-1；HaCaT；黄芩素；凋亡； 中图分类号：R751 Effects of Baicalein on Inducing Apoptosis and InhibitingProliferation in Human Cutaneous Squamous Cell Carcinoma Cell Line SCL-1 LU Jun, LIU Mei, HE Chundi (Department of Dermatology,No.1 Hospital of China Medical University Liaoning Shenyang 110001) Abstract: Objective: In this study, we used baicalein to deal with human skin squamous cell carcinoma cell line SCL-1 cells, and observed the effect of baicalein on the growth of human skin squamous cell carcinoma cells . METHODS:SCL-1 cells and HaCaT cells were cultured with methyl thiazolyl tetrazolium (MTT) assay to detect cell proliferation inhibition rate ,and cell morphology was observed under an inverted microscope, early cellular apoptosis was measured by flow cytometry , and morphological changes of apoptosis was observed with acridine orange staining. Results: pairwise comparisons of different time groups,the difference of effect of the treatment of baicalein after 48h 24h 12h was statistically significant, SCL-1 cells treated for 24h with 50μg/ml of baicalein solution, the apoptosis of SCL-1 cells appeared . Conclusion:baicalein has proliferation inhibition effect on SCL-1 cell,and it is more pronounced with the prolonged duration ,it also can induce apoptosis of SCL-1. The effect of Baicalein on HaCaT cell growth inhibition was not obvious; all-trans retinoic acid can also inhibit the proliferation of SCL-1 cells; a certain concentration of baica
