当归 当归 、红芪超滤膜提取物对小鼠机体及乳鼠心肌细胞辐射损伤保护作用的实验研究 PAGE PAGE 10 content of bone marrow cells Abstract two Objective: To observe whether Ultra-filtration Extract from Angelica sinensis and Radix Hedysari has the protective effect on myocardial radiation injury . Methods: Primary cultures of cardiac myocytes were prepared from ventricles of 1st-to-3rd-day-old Wistar rats and cultured for 3 to 4 days. Myocardial cells were randomly divided into normal control group (C), radiation injury group (RI), low-dose drug group (LD), middle dose group(MD) and high-dose drug group (HD). The modle of radiation injury was extablished through X-ray radiation in cultured cardiomyoctes of neonatal rats after 24 hours drug effects.Then Superoxide dismutase (SOD) was measured in all groups; Bcl-2 expression was observed by Immunohistochemistry. Results: We haved obtained a good isolation of cardiomyocytes,which with purity higher than 95%.Cultured for 3 to 4 days of myocardial cells in the implementation of radiation injury, SOD was significantly lower than normal (p 0.05); LD, MD, HD group were higher than injury group (p 0.05); Bcl-2 expression was upregulate compared to (RI) group,there was significant difference (p 0.05). Conclusion: (1) This study indicate that X-ray radiation can significantly result in radiation injury in cultured Cardiomyocytes of neonatal rat;(2)Ultra-filtration Extract from Angelica sinensis, Radix Hedysari (100,000 molecular weight) can reduce the radiation damage of myocardial cells.
