The expression characteristics and regulation of chitin synthase genes and hypothetical cuticular protein homologue gene in Bombyx moriAbstractInsect cuticular protein (ICP) and chitin are important components that affecting the mechanical properties of the epidermis, and they have a significant influence on growth and development of insect. So ICP and chitin are one of the important targets for developing specific biological insecticide. Based on the work of our laboratory, we use RT-PCR, qRT-PCR, RNAi, Western blotting and fluorescent staining of tissue section techniques to study the biological function of two chitin synthase genes in Bombyx mori (ChsA and ChsB), and its relationship with the regulation of hormones and inhibitors. We also research the hypothetical cuticular protein homologue (CPH-like) gene of Bombyx mori, and its role in midgut tissue formation and repair process. The results are as follows:The expression of ChsA and ChsB genes are regulated by exogenous 20E and JHAThe hormone exposure experiment showed the impact of exogenous 20E and JHA on ChsA and ChsB genes expression depended on the relative levels of endogenous hormone, rather than the regulation of a single molting hormone (MH) or juvenile hormone (JH). The exogenous hormones significantly up-regulated the expression of ChsA and ChsB genes in low level of endogenous MH and high level of endogenous JH right after molting of 4th and 5th larvae.
