南京农业大学学报 　2002 , 25 (2) : 71～74 　 Journal of Nanjing Agricultural University 土壤食细菌线虫的分离和富集培养方法 李辉信 , 陈小云 , 胡锋 (南京农业大学资源与环境科学学院 , 江苏 南京 210095) 摘要 : 从小麦根际土中分离得到一种能取食 B1 和 B2 两种细菌的土壤食细菌线虫 , 并在马铃薯蔗糖培养基上富化培养成 功。经鉴定该线虫为原小杆线虫属 ( Protorhabditis sp. ) , B1 为假单胞菌属 ( Pseudomonas sp. ) , B2 为醋杆菌属 ( A cetobacter sp. ) 。用牛肉膏蛋白胨细菌培养基培养B1 和 B2 两种细菌时 , 细菌无多糖产生 , 线虫不能取食细菌而生长繁 殖 ; 当用马铃薯蔗糖真菌培养基培养B1 和 B2 两种细菌时 , 细菌分泌丰富的多糖 , 线虫生长繁殖较好。多糖是线虫生长 繁殖的重要外部环境。 关键词 : 土壤食细菌线虫 ; 富化培养 ; 多糖 中图分类号: S1541 　　　文献标识码 : A 　　　文章编号 : (2002) Isolation and enrichment culturing of a soil bacterialfeeding nematode LI Huixin , CHEN Xiaoyun , HU Feng (College of Resources and Environmental Sciences , Nanjing Agric Univ , Nanjing 210095 , China) Abstract : A soil bacterivorous nematode feeding on bacteria B1 and B2 was isolated from rhizosphere soil of wheat and cultured on potato sucrose agar medium (PSA) . The nematode was identified as Protorhabditis sp. ; B1 and B2 were identified as Pseudomonas sp. and A cetobacter sp. , respectively. B1 and B2 didn’t secrete polysaccharose on beef polypeptone agar medium (BPA) and the nematode didn’t grow on BPA medium with B1 and B2 , while B1 and B2 secreted polysaccharose on PSA medium. The nematode grew very well on PSA medium with B1 and B2. Polysaccharose constitutes an important external environment for nematode growth.
