二甲苯对小鼠肾组织损伤的研究
摘 要
本试验以小鼠为试验动物,通过腹腔注射二甲苯染毒建立试验模型,测定小鼠肾组织中超氧化物歧化酶(SOD)活力和丙二醛(MDA)含量,以及制作组织切片进行病理学观察,从而分析评价二甲苯对小鼠的损伤程度。将小鼠随机分为Ⅰ、Ⅱ、Ⅲ、Ⅳ组,Ⅰ组为对照组(注射生理盐水),其他三组分别用不同浓度的二甲苯(用药剂量分别为0.125 ml/kg、0.25 ml/kg、0.5 ml/kg)经腹腔注射小鼠染毒15天,15天后处死、剖解小鼠,观察肾组织的大体变化并制作肾组织切片。使用SOD、MDA试剂盒测定不同试验组小鼠的肾组织匀浆中SOD的活力和MDA的含量。结果显示,Ⅳ组染毒小鼠的SOD活力较其他三组下降极显著(P0.01),而MDA的含量则增加极显著(P0.01);Ⅲ组和Ⅱ组的差异不显著,但相较于对照组差异显著(P0.05)。病理学变化:Ⅱ组肾组织肾近曲小管细胞之间边界模糊不清,但病变不是很明显;Ⅲ组肾组织肾近曲小管细胞之间边界模糊不清,细胞排列紊乱,脱落到管腔中,相对于Ⅱ组病变较明显;Ⅳ组见皮质区部分近端肾小管上皮细胞刷状缘脱落,有部分出现肾小球破裂、出血,部分近髓处小管上皮细胞肿胀,伴有空泡变性。

XYLENE ON RENAL INJURY IN MICE
ABSTRACT
In this experiment, mice as experimental animals exposed by intraperitoneal injection of xylene to build a test model, kidney tissue of mice, superoxide dismutase (SOD) activity and malondialdehyde (MDA) content, and the production of tissue in the disease Science observation, analysis and evaluation of xylene to injury in mice.
