齐 齐 哈 尔 大 学 毕业设计(论文) 题 目 香豆素衍生物荧光分子探针的合成及性能研究 学 院 化学与化学工程学院 专业班级 学生姓名 指导教师 成 绩 2012年6月12日 摘 要 以香豆素-3-甲酸为原料,经氯乙酰化得中间体香豆素-3-甲酰氯,进一步与2-吡啶-8-氨基喹啉反应,得荧光分子探针(SHK-1);2-吡啶-8-氨基喹啉经氯乙酰化得中间体,进一步与无水哌嗪反应,最后与香豆素-3-甲酰氯合成得SHK-3。上述产物的结构经1H-NMR表征,均为目标产物。 结果表明,关键词:香豆素;荧光分子探针;Cu2+ Abstract Coumarin-3-methyl chloride was synthesized using the raw marterial coumarin-3-formic acid through chloroacetylation. which were used as mediums of synthesizing complicated fluorescent probes. Furtherly, used coumarin-3-methyl chloride and 2-pyridine-8-aminoquinoline, fluorescent molecular probes SHK-1 was designed and synthesized through reaction. SHK-2 was synthesized using the coumarin-3-methyl chloride and excess ethylenediamine.. 2-pyridine-8-aminoquinoline was chloroacetylated using the raw marterial , which is the mediums of synthesizing complicated fluorescent probes. And then further reaction with anhydrous piperazine, finally the SHK-3 was synthesized through coumarin-3-methyl chloride. The structure was confirmed by 1H-NMR characterization of the above products, are the target product. After initial screening, fluorescent molecular probes of the SHK-1 was studied in the recognition performance of the metal ions in the neutral buffer solution. The results show that SHK-1 can be selective fluorescence quenching identification of Cu2+,Fe3+.After complexation with Fe3+, at the wavelength of 351nm, the fluorescence peak intensity quenching of 31%; After complexation with Cu2+, at the wavelength of the 474 nm, The fluorescence intensity is almost complete quenching, the quenching ratio of 97%. Quantitative analysis showed that SHK-1 formed a 1:1 complex with Cu2+. Metal ion competition experiments show that the detection of SHK-1 for Cu2+ did not been interfered for the common metal io
