益气补肾中药对自然流产模型小鼠NK细胞的影响

[摘要]:目的:观察益气补肾中药对自然流产模型小鼠脾脏、母胎界面组织NK细胞表达的影响。方法:以CBA/J♀×DBA/2♂为自然流产模型,妊娠第0天开始灌服益气补肾中药,并与环孢素A对照,孕第9天、14天流式细胞术分析脾脏、母胎界面组织NK细胞的表达情况。结果:孕9天时各用药组NK细胞在脾脏及母胎界面的表达较阴性对照组明显降低(P0.01 或P0.05);孕14天时各用药组NK细胞在脾脏的表达较阴性对照组明显降低(P0.05);孕14天时中西医结合组NK细胞在母胎界面的表达较阴性对照组明显降低(P0.05),而中药组、环孢素A组与阴性对照组比较无明显差异(P0.05)。结论:益气补肾中药可以下调自然流产模型小鼠孕早期及孕中晚期脾脏NK细胞表达及孕早期母胎界面NK细胞表达;益气补肾中药与环孢素A协同可以下调自然流产模型小鼠孕中晚期母胎界面NK细胞表达,具有一定的免疫调节作用。

The Effect of the Traditional Chinese Medicine with Function of Invigorating Qi and Tonifying Kidney on Natural Killer Cells in Abortion-prone Mice

Abstract Objective: To observe the effect of the traditional Chinese medicine (TCM) with function of invigorating Qi and tonifying kidney on the expression of natural killer (NK) cells at spleen and maternal-fetal interface in abortion-prone mice during pregnancy. Methods: CBA female mice were mated to DBA /2 male mice as an abortion-prone model. Drenched the TCM decoction we’ve made with function of invigorating Qi and tonifying kidney everyday since day 0 of gestation; And compared with Cyclosporin A. On the 9th and 14th day of gestation, the mice were killed in batches and their splenic and decidual tissues were made into single cell suspension to detect the expression of NK cells by FCM. Results: on the 9th day of gestation, the expressions of NK cells at spleen and decidual tissues in all of the therapeutic groups were reduced obviously, comparing with the result in negative control group (P0.01~0.05); on the 14th day of gestation, the expressions of NK cells at spleen in all of the therapeutic groups were reduced obviously, comparing with the result in negative control group(P0.05), and the expressions of NK cells at maternal-fetal interfac
