Axin蛋白表达及肝细胞癌发生及预后关系探究 　 作者：董承伟　曾勇　丁怡　庄冠一　李森 【摘要】 目的：明确Axin蛋白在正常肝组织、肝细胞癌组织、癌旁组织中的表达，探讨Axin蛋白表达与肝细胞癌的发生及与预后影响因素间的关系。方法：采用免疫组织化学方法(SABC法)检测正常肝组织(15例)、肝细胞癌组织(40例)及其癌旁组织(21例)中Axin蛋白的表达及定位。结果：正常肝组织、肝细胞癌组织及癌旁组织中均有Axin蛋白表达，其阳性表达率分别为92.9%、57.5%和90.5%。Axin蛋白表达于阳性细胞胞质中;正常肝组织与癌旁组织中Axin蛋白的表达差异无统计学意义(Pgt;0.05);正常肝组织和癌旁组织中Axin蛋白表达与癌组织比较，差异均有统计学意义(Plt;0.05);Axin的表达与肝细胞癌患者性别、年龄、肿瘤大小、AFP表达水平无关(Pgt;0.05)，而与临床病理分期、门静脉癌栓形成相关(Plt;0.05)。结论：Axin蛋白表达减弱可能参与肝细胞癌的发生;Axin蛋白表达水平可作为肝细胞癌患者诊断和预后判断的一项指标。 【关键词】 癌，肝细胞·Axin蛋白·免疫组织化学 　　【ABSTRACT】 Objective: To detect the expression and distribution of Axin in the normal liver tissue, paracarcinomatous tissue and primary hepatocellular carcinoma tissue and investigate the relationship between Axin expression and the genesis, and prognosisof HCC. Methods: The expression of Axin protein was evaluated in paraffin embedded tissues by immunohistochemical techniques (SABC) in 40 case of HCC, 21cases of paracarcinomatous tissue and 14 cases of normal liver tissue.And 5 case of normal stomach tissue and 5 cases of gastric carcinoma tissue were evaluated as a positive control. Results: The expression of Axin protein was observed in HCC tissues, paracarcinomatous tissues and normal liver tissues. The positive expressions of Axin were 57.5%、90.5%、92.9% respectively. The brown granules of positive cell were mainly distributed in the cytoplasm. No significant difference in the expression of Axin was found between normal liver tissues and paracarcinomatous tissue(Pgt;0.05), but not between the paracarcinomatous tissue and HCC(Plt;0.05). Axin protein expression were not related to age, sex, tumor size and the expression of AFP, but were significantly correlated with portal vein invasion and clinical pathological stage. In normal stomach tissue, the expression of Axin is strong in the stem cells and weak in the mature cells. But its expression in normal liver tissue is well-proportioned.
