T细胞因子3在非小细胞肺癌中的表达研究 闵利1，刘煜亮1，徐瑜2，李万成3，陈虹 1.重庆医科大学附属第一医院呼吸与危重症医学科，重庆 400016；2.第三军医大学新桥医院呼吸内科，重庆 400037；3.成都医学院第一附属医院呼吸内科，成都 610500 [摘要] 目的: 研究T细胞因子3（T cell factor 3,TCF-3）在非小细胞肺癌中的表达及分布情况。方法: 采用50对人非小细胞肺癌组织标本及其正常癌旁组织对照，进行免疫组化检测TCF-3蛋白在肺癌组织及正常肺组织中的表达及分布情况；采用5种人非小细胞肺癌细胞系及2种正常人支气管上皮细胞系进行实时荧光定量PCR及Western blot 检测肺癌细胞系中TCF-3在mRNA和蛋白水平的表达情况。 结果:(1) TCF-3在肺癌组织中表达，表达率为48%（24/50），在正常肺组织中未见表达。TCF-3在肺癌细胞胞核与胞质同表达，以细胞核表达为主。(2)在7种细胞系中TCF-3在A549、H520、HBE、BEP-2D四种细胞系中mRNA及蛋白水平均有表达。结论: TCF-3在肺癌组织及细胞系中均表达，TCF-3蛋白在细胞核与细胞质同表达，但以细胞核表达为主。 关键词：T细胞因子3/TCF-3；非小细胞肺癌；Wnt通路 中图分类号：R734.2 The expressions of T cell factor 3 in non-small cell lung cancer Min Li1, Liu Yuliang 1 ，Xu Yu2, Li Wancheng3, Chen Hong 1 1.Department of Respiratory and Critical CareMedicine, the First Affiliated Hospital of Chongqing Medical University, Chongqing 400016, China; 2. Department of Respiratory Medicine，The First Affiliated Hospital of Chengdu Medical College, Chengdu 610500, China [ABSTRACT] Objective: To study the expression and distribution of T cell factor-3 (TCF-3) in non-small cell lung cancer (NSCLC). Methods: TCF-3 expression was examined in 50 non-small cell lung cancer specimens and 50 corresponding normal lung specimens using immunohistochemistry(S-P method). The mRNA level of TCF-3 in 5 NSCLC cell lines ( A549, H460, H226, H520, SK-MES-1 ) and 2 human normal bronchiepithelium cell lines ( HBE and BEP-2D ) were detected by real-time fluorogenic quantitative PCR. The protein level of TCF-3 gene was detected by Western blotting. Results:(1) Among 50 lung cancer specimens, 24 samples showed high expression of TCF-3 (48%), including 22 samples showed TCF-3 expression both in cytoplasm and nuclei ,TCF-3 were not detected in 50 corresponding normal lung specimens.（2）The mRNA and protein expression level of TCF-3 was significantly higher in A549,H520,HBE,BEP-2D than its
