第29卷 第12期 传 感 技 术 学 报 Vol29 No.12 2016年12月 CHINESEJOURNALOFSENSORSANDACTUATORS Dec. 2016 AStudyonMouseGermCell-BasedSensor fortheDetectionofBitterness TIANYulan，SUKaiqi，QIUXianxin，FANGJiaru，QINZheng，LIRong，WANGPing* （BiosensorNationalSpecialLaboratory，KeyLaboratoryforBiomedicalEngineering，MinistryofEducation， DepartmentofBiomedicalEngineering，ZhejiangUniversity，Hangzhou3 0027，China） Abstract：Inthisstudy，thefeasibilityofutilizingmalemousegermcellsintestisandelectricalcell-substrateimped⁃ ancesensor（ECIS）wasexploredtobuildacell-basedbitterbiosensor，whichcansensitivelyandspecificallyrespond tobittercompounds.MalemousegermcellsexpresslotsofbitterreceptorT2Rs（Gproteincoupledreceptors）which cansensitivelyandspecificallyrespondtobittercompounds，whenactivebyligandswillaffectcellmorphologyina specificmanner.Thebestcelldensitywasexplored；cell-impedanceresponseprofilesofgermcellstotwobittercom⁃ poundswereinvestigatedbyanalyzingtheresponseintensityundervariousconcentrations，Thelineardetectionrange ofPTCis10μmol/L~200μmol/L，thedetectionlimitis4μmol/L；thelineardetectionrangeofquinineis62.5μmol/L~ 1000μmol/L，thedetectionlimitis4μmol/L； Furthermore，impedanceresponsestofivebasictasteswereexam⁃ inedtoevaluatethespecificityofcell-basedbitterbiosensorinbitterdetection.Theresultsrevealedthatthishybrid bitterbiosensorcouldrespondtothosebittercompoundsinadose-dependentmanner.Anditcouldrespondtobitter compoundsspecifically.Thisbiosensormayprovideapromisingapproachfordetectingvariousbittercompounds.
