Ticlopidine and clopidogrel inhibit platelet activation at an early stage by irreversibly and selectively binding to the adenosine diphosphate receptor. This inhibits the activation of the glycoprotein IIb/IIIa receptor. Dipyridamole inhibits phosphodiesterase, thereby reducing the breakdown of cyclic adenosine monophosphate and enhancing the production of prostaglandin I2, and leading to vasodilation and the inhibition of platelet aggregation. Abciximab, eptifibatide, and tirofiban block the final stage of platelet aggregation by preventing the binding of fibrinogen. In theory, as antiplatelet agents have different mechanisms of action, they can be used in combination with ASA to achieve additional antiplatelet effects.1 However, many of these additional benefits tend to be small, outweighed by the increased risk of bleeding, and usually not cost-effective. 1.
