 猪卵泡闭锁程度的判定方法及比较研究# 张金璧，潘增祥，林飞，马雪山，李齐发，刘红林** （南京农业大学动物科技学院，南京 210095） 5 10 15 20 25 30 35 40 摘要：在大多数哺乳动物中，雌性个体在新生时就已拥有最大数目的原始卵泡储备库，而随 着动物的发育和成熟，绝大多数卵泡发生了闭锁和退化，直至最终的完全耗竭。为了细致地 研究卵泡闭锁机制，对卵泡的闭锁程序进行准确合理的判断必不可少。本实验以猪卵泡为实 验材料，在总结各种分类方法的同时，通过具体的实验分析了几种可持续跟踪研究的分类方 法（外观形态，颗粒细胞密度，P4 和 E2 的比值）间的关系。本研究经过对 239 个卵泡的分 离和检测，以及对在不同闭锁阶段各 20 个，共 60 个卵泡在各个判断指标上都达到一致的卵 泡的分析和比较，检验各判定指标与猪卵泡闭锁的关系，结合表观形态，形成了合理的判别 标准：颗粒细胞密度2500 个/μL、2500-10000 个/μL 及10000 个/μL 分为健康、早期闭 锁及晚期闭锁卵泡，而相应的 P4 和 E2 比值分别为 P4/E25、5P4/E220 及 P4/E220。 关键词：猪；卵巢；卵泡；闭锁；闭锁程度 中图分类号：S813 Study on the approaches to determine the extent of follicular atresia in pigs and comparison thereof ZHANG Jinbi, PAN Zengxiang, LIN Fei, MA Xueshan, LI Qifa, LIU Honglin (College of Animal Science and Technology, Nanjing Agricultural University, NanJing 210095) Abstract: In many mammalians, the ovary is endowed at birth with a fixed primordial follicles repertory, which steadily dwindles throughout life as a result of atresia and recruitment towards ovulation till the ultimate exhaustion. In order to explore the detailed mechanism of follicular atresia, it is indispensable to accurately determine and describe the atresia procedures of follicles. At present, there is no a consistent method for deciding the extent of follicular atresia, this has gave rise to some obstruction in academic exchange or studies as lacking of a unified standard. In this study, using porcine follicles as experimental material, we comprehensively summarized and compared a variety of methods for atresia follicle classification, analyzed the relationship between several continuable and trackable indes (morphology, density of granulosa cells, P4 and E2 ratio) by specific assay. In this study, 239 follicles were separated and analyzed, and a total of 60 follicles in different stages of atresia (20 in each stage) were picked out, which all achieved the consistency of the above three continuable and trackable indes.
